CD4+ T cells from patients with primary biliary cholangitis show T cell activation and differentially expressed T cell receptor repertoires
Hepatol Res. 2019 Jun;49(6):653-662.

Our TCR repertoire analysis was used in the study of primary biliary cholangitis, an autoimmune disease. As a result, it has been reported that there are different TCR repertoires in the patient group and the healthy subject group in the CD4+ T-cell population. In addition, the authors suggest that the CD4+ T-cell population in the patient population may be associated with high expression of the N-Ras gene.

Brain regulatory T cells suppress astrogliosis and potentiate neurological recovery

Our TCR repertoire analysis was used in a study using a cerebral infarction model mouse. As a result of performing TCR repertoire analysis, it is reported that the accumulated T-reg cells are composed of a population different from that of the periphery. As a result, it is suggested that T-reg cells accumulating at the infarct lesion site may control nerve repair.

Our repertoire analysis was used in nickel allergy research. The authors created a nickel allergy mouse model and reported differences in TCR repertoire in lesions. These results suggest that activation of NKT cells and unique TCR sets may be induced in nickel allergy.

Epstein-Barr virus-related diffuse large B-cell lymphoma in mogamulizumab-treated adult T-cell leukemia with incomplete T-cell reconstitution

Our repertoire analysis was used in the study of leukemia. The authors performed a TCR repertoire analysis of adult T-cell leukemia (ATL) that developed EBV-related B-cell lymphoma (DLBCL) after administration of anti-CCR4 antibody (mogamulizumab). As a result, the authors reported the identification of ATL clones after treatment, decreased peripheral blood diversity, and the presence of ATL clones in skin rashes. These results suggest a link between immune abnormalities in T cells and the development of DLBCL.

Induction of oligoclonal CD8 T cell responses against pulmonary metastatic cancer by a phospholipid-conjugated TLR7 agonist
PNAS. 2018 Jul 17;115(29):E6836-E6844.

Our repertoire analysis was used in the study of tumor immunity. The authors report that a single dose of 1V270, a phospholipid-bound Toll-like receptor 7 (TLR7) agonist, suppresses the growth of advanced lung metastatic tumors. Furthermore, it is mentioned that the induction of tumor-specific CD8+ T cells is common in the lung and mediastinal lymph nodes. These results suggest that systemic 1V270 therapy induces tumor-specific cytotoxic T-cell responses to lung metastatic cancer, and that repertoire analysis is useful for monitoring therapeutic responses.

Mitophagy in Intestinal Epithelial Cells Triggers Adaptive Immunity during Tumorigenesis
Cell. 2018 Jun 28;174(1):88-101.e16.

Our repertoire analysis was used in the study of tumor immunity. The authors used a TCR repertoire analysis to evaluate the association of MHC class I with the function of mitochondria and lysosomes in intestinal epithelial cells. As a result, the authors identified the diversity and individual clones of CD8+ T cells in the mucosa of genetically modified mice. These results suggest that treatments that induce mitophagy in intestinal epithelial cells and induce lysosomal membrane permeability may improve the balance of antitumor immunity against colorectal cancer.

Quantitative T-cell repertoire analysis of peripheral blood mononuclear cells from lung cancer patients following long-term cancer peptide vaccination

Our repertoire analysis was used in the study of peptide vaccine therapy. The authors performed a repertoire analysis of peripheral blood and tetramer-sorted peptide-specific T cells in lung cancer patients receiving peptide vaccine therapy and followed the changes over time for a long period. As a result, the authors reported that the clones were replaced depending on the treatment time, suggesting that multiple T cells respond to the treatment.
